Specific to eQuest, are you talking about subdividing a room into multiple zones? OR talking about applying a multiplier at the space input to have multiple of the same space? Needing some context to address

actually, I want to separate one specific room to different thermal zone. this means that I want to make a more accurate model when I calculate and talk about the room. because as we know each wall of a room has a different condition. so I want to consider this different thermal condition in each part of my room to make an accurate model.

You can use different thermal zones in one physical room. What you need to do is to add air walls for each zones.

I have not used eQUEST that much, but in OpenStudio you can group spaces under one zone using the SketchUp plugin. Check this Video (minute 3:00)
