running simulation for one day only

Can the simulation be run for only one day? And if it so, does the weather file need to contain only one day, or OpenStudio knows how to pick this day from the EPW file? And if I'm running for one day only, can I insert the Design Days information manually for this day only? Thanks very much.

Yes. Use the run period selectors on the simulation settings tab shown here:

The simulation will pick the correct weather from the EPW file. Design days are used to size equipment for extreme conditions - that's distinct from the days the simulation is being performed. I would recommend using the extreme days from the DDY file as opposed to sizing the system using the conditions on that single day you are simulating.
