I want to know how can I set the "Tolerance temperature" in E+ which is the temperature control. The situation can be describe that I want to use AC when the room temperature is higher than 29 ℃,but if I use AC the cooling setpoint is 26 ℃.

So which temperature should I fill in HVAC Templates:Themostat Constant Cooling Setpoint ? And if I can set the two schedule to meet the situation that when the room temperature lower than 29 ℃ I close the AC although the operation schedule is on.

Could have been helpful to share what type of AC system you plan on using, but either way I think you can do the following:

This would look like the following, making sure you replace the 'Sensor Node' by a valid Outside Air node

AvailabilityManager:HighTemperatureTurnOn,
  AC HighTemp Turn On Availability Manager, !- Name
  Outside Air Inlet Node, !- Sensor Node
  29.0;   !- Temperature {C}
