Infiltration is typically defined from the Space Types Tab (5th from top). You'll see these on the Spaces Tab (7th from top) under the Loads subtab. These will typically be in green text to designate that the values were inherited from the Space Type definition. You can choose to redefine the values used in the Space Type Tabs.

Infiltration is treated as a load associated with a physical space. Thermal zones can have a 1-to-1 relationship with spaces, but that's not a requirement (e.g. a thermal zone can span multiple physical spaces).

Yes. Specify the infiltration as a space type definition -> Assign the space type definition to a physical space -> Zone your building. It may seem like an extra step, but it gives you flexibility in evaluating different zoning options for your building - and there's a user script in the SketchUp Plug-In that will automatically create a 1-to-1 mapping between spaces and zones for you if you want to zone your building that way. (I don't recall if that's been converted into a standalone OpenStudio Measure or not.)
