I'm modeling a building with DesinBuilder for thermal comfort testing with EnergyPlus. In these tests I need to compare thermal comfort through PMV and operating temperature for different window and door opening levels. I want to test the efficiency of natural ventilation, but I'm having trouble inserting objects for opening manipulation. Could someone help me by telling me which objects I need to insert to get the data from these aperture variations?

I would recommend starting the DesignBuilder tutorial for Scheduled natural ventilation and infiltration. You can also get more information about these DesignBuilder inputs in the DesignBuilder help guide section for external glazing window operation and free aperture.

To review outputs for PMV and operating temperature in DesignBuilder, you will need to enable those in the Output options when you run a simulation.
